What is Liraglutide?

Thirteen hours is long enough for daily dosing and nothing more. That is the single most important fact about this compound and it is what separates it from the weekly ladder that semaglutide, tirzepatide, and retatrutide sit on. Liraglutide escalates 0.6 -> 1.2 -> 1.8 -> 2.4 -> 3.0mg, one step per week, injected every single day. It also predates all of them, so the literature is deep human clinical data rather than the animal-and-inference picture most of this catalogue runs on. Older search traffic still finds it under its trade names, Saxenda and Victoza; the molecule is the same liraglutide either way.
For a 5mg vial the daily cadence has a blunt consequence: at the 3.0mg maintenance step, 5mg is under two days of material. This vial size fits the bottom of the escalation ladder, not the top.

Human pharmacokinetics are unusually well characterised for a peptide in this catalogue: Tmax 8-12 hours, absolute bioavailability around 55%, plasma protein binding over 98%, apparent volume of distribution around 13L, apparent clearance around 1.2 L/h. Degradation happens through general endogenous protein catabolism rather than a single clearance organ, with only trace liraglutide-related material recovered in urine and faeces.
The efficacy literature is human and large. The SCALE weight-management programme ran 3.0mg daily against placebo for 56 weeks in over 3,700 participants; LEADER ran cardiovascular outcomes in type 2 diabetes at up to 1.8mg. Read those as clinical pharmacology on the pharmaceutical article, not as claims about research-grade material.
