SNAP-8 vs Argireline: Vial Sizes and Dosing

TL;DR: Argireline (Acetyl Hexapeptide-8, 889 Da) and SNAP-8 (Acetyl Octapeptide-3, 1075 Da) are the same SNAP-25 mimetic idea with two extra residues on the octapeptide. They get conflated because they arrive in three different formats: a 10mg lyophilised vial, bulk powder, and a pre-diluted cosmetic trade solution that is only 0.05% peptide. Sort the presentation first, then the maths is trivial — 10mg in 2mL is 5 mg/mL, 50 mcg per unit. The Argireline calculator handles the conversions.

Three presentations, one molecule family

The single biggest source of confusion is that “Argireline” and “SNAP-8” both name a peptide and a commercial preparation, and the two differ by roughly three orders of magnitude in strength.

  • The 10mg lyophilised vial. A bench format: freeze-dried peptide cake, reconstituted with bacteriostatic water. This is what Argireline 10mg and SNAP-8 10mg refer to.
  • Bulk powder. Sold in 100mg to 500mg quantities (see SNAP-8 500mg) because formulation work at any real scale burns through milligrams fast.
  • The cosmetic trade solution. Supplied as an aqueous dilution — the SNAP-8 solution sold to formulators is 0.05% acetyl octapeptide-3, with a suggested incorporation rate of 3-10% into a finished base. The Argireline equivalent carries an INCI of Water, Acetyl Hexapeptide-8, Acetic Acid, Phenoxyethanol, Potassium Sorbate, with a 2-10% use level.

Sequence-wise, Argireline is Ac-Glu-Glu-Met-Gln-Arg-Arg-NH2 and SNAP-8 is that same N-terminal SNAP-25 fragment with Ala-Asp added. Everything downstream — solubility, storage, reconstitution behaviour — is near-identical. The differences that matter for dosing are molecular weight and how each one is packaged.

The 10% trap

When someone says a serum is “10% Argireline,” they almost always mean 10% of the trade solution, not 10% peptide. Run the arithmetic:

0.05% peptide in solution × 10% incorporation = 0.005% peptide in the finished product.

That is 50 mcg of peptide per gram of cream — 50 ppm. At the low end of the range (3%) it is 0.0015%, or 15 ppm. Meanwhile the founding clinical paper, Blanes-Mira et al. (2002), describes an oil/water emulsion “containing 10% of the hexapeptide” that reduced wrinkle depth by up to 30% over 30 days in a small volunteer group. If you read that literally as 10% peptide by weight, it is 2,000 times the concentration in a typical off-the-shelf product carrying the same “10%” number.

This is also why the 10mg vial and the bulk powder exist as separate SKUs. One is for characterisation and small measured aliquots; the other is for actually making something.

Reconstituting the 10mg vials

Both peptides are strongly hydrophilic and dissolve readily in bacteriostatic water. The formula is the same one every lyophilised peptide uses:

mg/mL = vial strength (mg) ÷ BAC water added (mL)

At the standard 2mL fill on a 10mg vial:

  • Concentration: 10 ÷ 2 = 5 mg/mL = 5000 mcg/mL
  • On a U-100 syringe, 1 mL = 100 units, so 5000 ÷ 100 = 50 mcg per unit
  • A 1 mg aliquot: 1000 ÷ 50 = 20 units (0.20 mL), giving 10 aliquots per vial

Halve the water to 1mL and you get 10 mg/mL — 100 mcg per unit, and that same 1 mg aliquot becomes 10 units. Nothing about the peptide changed; you only changed the ruler. Whether you are working out an Argireline dosificación or a SNAP-8 Dosierung, the division is identical for both compounds, because they ship at the same nominal vial strength.

Worked example: one vial into a cream base

Say you want to prepare a 30 g base at 0.005% peptide — the concentration a commercial “10%” product actually delivers.

  • Peptide needed: 30 g × 0.00005 = 0.0015 g = 1.5 mg = 1500 mcg
  • At 50 mcg/unit: 1500 ÷ 50 = 30 units, or 0.30 mL of reconstituted solution

Which means one 10mg vial covers a lot of ground at that strength. Divide 10 mg by 0.00005 and you get 200 g of finished preparation — nearly seven 30 g jars. Step the target up and the yield collapses fast:

That last figure is the punchline. Matching the 10%-by-weight preparation from the literature in a 30 g base needs 3 g of peptide — 300 vials of the 10mg size, or six of the 500mg bulk vials. That single calculation explains the entire product catalogue: nobody formulates from 10mg vials, and nobody characterises from 500mg drums.

Weight versus moles: 889 against 1075

Here is where the two siblings genuinely diverge on paper. SNAP-8 is 21% heavier per molecule, so equal masses are not equal molar doses.

  • 10 mg Argireline ÷ 889 g/mol = 11.25 µmol
  • 10 mg SNAP-8 ÷ 1075 g/mol = 9.30 µmol

A 10mg SNAP-8 vial therefore holds about 17% fewer molecules than a 10mg Argireline vial. To match on a molar basis you would need 10 × (1075 ÷ 889) = 12.1 mg of SNAP-8 — so a 1% Argireline preparation is molar-equivalent to roughly a 1.21% SNAP-8 preparation.

Whether that correction is meaningful is an open question, because head-to-head comparisons of the two are generally reported on a weight basis by the ingredient supplier rather than in independent literature. But if you are running a comparaison SNAP-8 Argireline on the bench, weight-matching and molar-matching are two different experiments and worth stating explicitly.

What the evidence actually supports

Being straight about this: the two compounds have very different evidence bases, and neither is strong.

Argireline has the better-documented side. Blanes-Mira et al. (2002) reported the original 10% emulsion result and showed the peptide inhibits neurotransmitter release with potency comparable to botulinum neurotoxin A but far lower efficacy. A 2025 review in Int J Mol Sci pulled the permeability literature together and found it contradictory — one study reporting about 30% of applied peptide crossing a membrane in two hours, another finding only 0.22% penetrating the stratum corneum over 24 hours — and concluded that low skin penetration limits bioavailability, with the underlying mechanisms “incompletely understood.”

SNAP-8 has almost nothing peer-reviewed on efficacy. The widely repeated “63% wrinkle reduction” and “30% better than Argireline” figures trace to supplier-sponsored testing, not to independent published trials. The published SNAP-8 literature is mostly analytical: Ji et al. (2020) developed an LC-MS/MS method for quantifying acetyl octapeptide-3, reaching a limit of quantification of 0.0125 ng/mL and applying it to a microneedle patch. That is a methods paper, not an outcome paper.

Frequently Asked Questions

Is SNAP-8 stronger than Argireline?

Supplier testing claims roughly 30% greater wrinkle-depth reduction for SNAP-8 at matched concentrations, but that comparison has not been reproduced in independent peer-reviewed literature. On a molar basis SNAP-8 is actually 17% less concentrated at equal weight, since it is a heavier molecule. Treat the claim as unverified.

How much bacteriostatic water for a 10mg Argireline vial?

2mL is the standard, giving 5 mg/mL — 50 mcg per unit on a U-100 syringe. Use 1mL if you want fewer, more concentrated draws (100 mcg/unit). The peptide dissolves quickly either way; swirl gently rather than shaking. The calculator converts any volume you pick.

Does a 10% Argireline serum contain 10% peptide?

No. It contains 10% of a trade solution that is itself around 0.05% peptide, so the finished product is roughly 0.005% acetyl hexapeptide-8 — about 50 mcg per gram. The 10% figures quoted from clinical papers refer to a different denominator entirely.

Can I use SNAP-8 and Argireline together?

They target the same SNARE-complex mechanism, so combining them is closer to raising the total dose of one idea than to stacking two mechanisms. There is no published data on the combination. If you are comparing them on the bench, running them separately at molar-matched concentrations gives cleaner information.

How many doses are in a 10mg vial?

Depends entirely on what you are preparing. At the 0.005% level typical of finished cosmetic products, one 10mg vial yields about 200 g of preparation. Split into 1 mg aliquots at 5 mg/mL, it gives ten draws of 20 units each.

Why does SNAP-8 come in a 500mg vial when Argireline comes in 10mg?

Because the presentations serve different jobs. Matching a 10%-by-weight study preparation in a 30 g base needs 3 g of peptide — 300 of the small vials. Bulk sizes exist for formulation work; the 10mg vial exists for characterisation, stability checks and measured aliquots.
